Na een schipbreuk waarbij zijn vader omkomt, wordt miljardair Oliver Queen vijf jaar lang vermist en doodgewaand, voordat hij wordt teruggevonden op een afgelegen eiland in de Stille Oceaan. In zijn geliefde Starling City wordt hij met open armen verwelkomd door zijn toegewijde moeder Moira, zijn geliefde zusje Thea en zijn beste vriend Tommy. Al snel merken ze dat Oliver is veranderd door zijn beproeving op het eiland. Terwijl Oliver de waarheid over de man die hij is geworden geheim houdt, wil hij de fouten uit zijn jonge jaren goedmaken en probeert hij zich te verzoenen met zijn ex-vriendin, Laurel Lance.

Heimelijk creëert hij de persona van Arrow, een vigilante, om het onrecht dat zijn familie is aangedaan en de uitwassen van de samenleving te bestrijden en zijn geliefde stad Starling City weer in haar oude glorie te herstellen. Overdag speelt Oliver de rol van de rijke, zorgeloze playboy, die hij ooit was, bijgestaan door zijn toegewijde chauffeur/bodyguard, John Diggle, terwijl hij onder de dekking van de duisternis zijn geheime identiteit aanneemt. Laurels vader, rechercheur Quentin Lance, is vastbesloten om de onbekende vigilante te arresteren. Ondertussen wordt duidelijk, dat Olivers moeder, die inmiddels hertrouwd is met een zakenpartner van zijn vader, meer weet van de dodelijke schipbreuk en meedogenlozer is dan hij zich ooit had kunnen voorstellen.

The series follows Oliver Queen (Stephen Amell), billionaire playboy of Starling City, who spends five years stranded on an island following a shipwreck that claims the life of everyone else on board, including his father, Robert Queen, and Sara Lance, the sister of Oliver's girlfriend Laurel (Katie Cassidy), and with whom he was having an affair. Upon his return to Starling City, he is reunited with his mother, Moira (Susanna Thompson), her new husband, Walter Steele (Colin Salmon), the former CFO of his father's company who is now the CEO and his younger sister, Thea (Willa Holland). He is also greeted by his best friend, Tommy Merlyn (Colin Donnell), the son of successful businessman Malcolm Merlyn (John Barrowman). Oliver also tries to reconnect with ex-girlfriend Laurel, but she blames him for the death of her sister, Sara.

During the day, Oliver plays the billionaire playboy who owns and runs a nightclub; at night, he becomes a green-hooded vigilante, following through with his father's dying wishes to right the wrongs of the Queen family, fight the ills of society, and restore Starling City to its former glory. Oliver's vigilante persona becomes the focus of police detective Quentin Lance (Paul Blackthorne), father to Laurel and Sara, who is determined to arrest the vigilante and also blames Oliver for his daughter's death and his wife leaving him, though he remains unaware of his dual identity. Oliver is constantly flanked by a bodyguard, John Diggle (David Ramsey), who later joins him in his fight for justice. Moira is also hiding secrets: she knows that the family yacht was actually sabotaged.
